RESUMO

Objective To study the genotoxicities of raceanisodamine hydrochloride injection. Methods Bacterial reverse mutation test, in vitro Chromosomal aberration test and in vivo Micronucleus test were performed to investigate the genotoxicities of raceanisodamine hydrochloride injection. Results The Ames test showed that raceanisodamine hydrochloride injection did not increase mutagenicity for TA1535, TA102, TA100, TA98 and TA97 strains at the dosage of 0.5, 5, 50, 500, 5000 μg per plate under two parallel system conditions (±S9). Results of CA test indicated that there was no statistical difference between raceanisodamine hydrochloride injection groups (doses of 58.75,117.5 and 235.0 μg/ml) and the solvent control group under two parallel system conditions (±S9). In MNT test, with doses of 7.5, 15.0 and 30.0 mg/kg respectively, the micronucleus induction rate of bone marrow of ICR mice was not statistically significant (P>0.05) when compared with that of vehicle control group in all dose groups. Conclusion Under the conditions of these study, the results indicated that raceanisodamine hydrochloride injection had no mutagenicity to Salmonella typhimurium, had no aberration effect on the chromosome of mammalian cultured cells, and had no effect on inducing micronucleus of bone marrow polychromatic erythrocytes in ICR mouse. All test results showed that raceanisodamine hydrochloride injection had no potential carcinogenicities and genetic toxicities under the test conditions.

Objective:To investigate the prognostic significance of preoperative neutrophil-lymphocyte ratio(NLR)combined with tu-mor-associated neutrophils(TANs)in the stroma of gastric cancer tissues of patients.Methods:One hundred and twenty-six gastric cancer tissue samples from patients enrolled in Changhai hospital from June 2006 to May 2011 were divided into four groups accord-ing to NLRs in preoperative peripheral blood combined with high or low infiltration of neutrophils in gastric cancer tissues.The 5-year survival of the four groups was then compared,and their correlations with clinicopathologic features and prognosis were analyzed.Re-sults:High NLRs in peripheral blood combined with low infiltrating TANs in gastric tissues was associated with lower differential grade (P<0.001)and larger tumor size(P=0.026).Of the four groups,patients with high NLR in peripheral blood combined with low infiltrat-ing TANs in gastric cancer tissues demonstrated the lowest survival rates,whereas those with low NLR and high infiltrating TANs had highest survival rates, and this difference was statistically significant (P<0.05). Univariate and multivariate Cox regression analyses showed that high NLR in peripheral blood combined with low infiltrating TANs in gastric cancer tissues(P<0.05)was an independent factor indicating poor prognosis.Conclusions:NLR in preoperative peripheral blood combined with infiltrating TANs in gastric cancer tissues can be used as a prognostic indicator for patients with gastric cancer,especially high NLR in preoperative peripheral blood com-bined with low infiltrating TANs in gastric cancer tissue indicates poor prognosis.

RESUMO

Objective Serum pharmacochemistry was performed to screen the bioactive constituents of Resina Draconis.Methods Based on HPLC fingerprints of Resina Draconis,the migrating constituents absorbed into blood were determined by comparing the HPLC fingerprints of extraction of Resina Draconis(ERB),herb serum sample,and control serum sample,and with the help of LC-MS/MS.Results Six compounds absorbed into blood were detected,five of them were original in form which were 3,4′-dihydroxy-5-methoxystilbene,cochinchinenin B,4′-hydroxy-4,2′-dimethoxy-dihydrochalcone(a new compound),cochinchinenin A,and loureirin B,respectively.The other might be the original constituent or the metabolite.Conclusion The six constituents absorbed into blood are possible bioactive components of Resina Draconis in vivo.Further research will help clarify the bioactive constituents and mechanisms of Resina Draconis.

RESUMO

Objective In order to elucidate neurotoxic actions of toluene in the air of submarine, the effect of toluene on biochemical alteration in the brain was studied in rat. Methods Adult healthy male Sprague-Dawley (SD) rats were randomly grouped into five groups (blank control group, excipient group, low dose group, medium dose group and high dose group). Toluene-exposed rats were administered with toluene in different doses (once a time every other day). Rats of excipient group were given olive oil with the volume as same as toluene. Nothing was given to rats of blank control group. 30 days later, all rats were sacrificed and the hippocampus and cortex were harvested for determination of nitric oxide synthase (NOS), nitric oxide (NO), malondialdehyde (MDA), Na~+-K~+-ATPase, reactive oxygen substances(ROS) and glutamic acid levels. Results NO content and NOS activity of rat's hippocampus in high dose group increased significantly compared with that of control group, and NO content was significantly positively correlated to NOS activity (r=0.7254, P
